(AP) ? The former graduate student accused in the deadly Colorado movie theater shooting was being treated by a psychiatrist at the university where he studied, the first indication that he may have sought help before the rampage that killed 12 people and wounded 58.

Attorneys for James Holmes, 24, made the disclosure in a court motion Friday as they sought to discover the source of leaks to some media outlets that he sent the psychiatrist a package containing a notebook with descriptions of an attack.

The motion said the leaks jeopardized Holmes' right to a fair trial and violated a judge's gag order.

Holmes' lawyers added that the package contained communications between Holmes and his psychiatrist that should be shielded from public view. The document describes Holmes as a "psychiatric patient" of Dr. Lynne Fenton.

The motion did not reveal when Holmes began seeing Fenton or whether he was being treated for a mental illness. Legal analysts expect Holmes' attorneys to use an insanity defense at trial. Holmes is scheduled to be arraigned Monday. A hearing on the new defense motion also is scheduled that day.

Calls to Holmes' lawyer and the state public defender's office were not immediately returned, nor was a message left with Fenton's office. A spokeswoman for the Arapahoe County prosecutor's office declined comment.

The University of Colorado's website identified Fenton as the medical director of the school's Student Mental Health Services. An online resume stated that she sees 10 to 15 graduate students a week for medication and psychotherapy, as well as 5 to 10 patients in her general practice as a psychiatrist. Schizophrenia was listed as one of her research interests.

Fenton was disciplined by the Colorado Medical Board in 2004 for prescribing herself Xanax while her mother was dying, state records show. She also was disciplined for prescribing the sleep aid Ambien and the allergy medicine Claritin for her husband, and painkillers for an employee who suffered from chronic headaches.

Fenton worked for the U.S. Air Force in Texas as an acupuncturist before joining the University of Colorado in 2005.

A 1998 Denver Post article quotes a Colorado acupuncturist named Lynne Fenton discussing how acupuncture could be used to enhance women's busts.

In the week since the July 20 attack at an Aurora movie theater, few details have emerged about Holmes' life ? especially the year he spent as a graduate student in Colorado. Holmes enrolled in a doctoral program in neuroscience at the University of Colorado-Denver Anschutz medical campus in June 2011. He left without explanation in June.

University officials have refused to disclose much more about Holmes, citing an order from the judge barring them from releasing information that would "impede an ongoing investigation." Staff, professors and classmates have been mum about Holmes' life at the school.

Holmes' appearance at his first court hearing on Monday stunned the victims' families and fueled speculation about the state of his mental health. His hair dyed a shocking comic-book shade of orange-red, he looked sleepy and, at times, inattentive.

Prosecutors said they didn't know if he was being medicated. Friday's defense motion, however, was the first confirmation from the defense that Holmes was seeing a psychiatrist and that he had sent a package to the doctor.

The package was seized by authorities on Monday after it was discovered in the mailroom at the university. It's unclear if it was sent before the attack at the July 20 midnight showing of "The Dark Knight Rises" that left 12 dead and dozens of others injured.

Authorities said Holmes legally purchased four guns before the attack at Denver-area sporting goods stores ? a semiautomatic rifle, a shotgun and two pistols. To buy the guns, Holmes had to pass background checks that can take as little as 20 minutes in Colorado.

Federal law bars from purchasing firearms people who have been found mentally defective by a judge or who have been committed to a mental health institution, said Benjamin Van Houten of the Law Center to Prevent Gun Violence.

States can impose additional restrictions, but Colorado does not.

"That is why it's important for states to look at going beyond federal law in this area," Van Houten said.

At a press conference earlier this week, university officials acknowledged that students in the neuroscience program are carefully monitored. They said a graduate student experiencing problems would normally be referred to student support services.

On Friday, 21 news organizations, including The Associated Press, filed a motion asking the judge to reverse his order sealing court records in the case. The news organizations claimed the order means even a complete list of court filings is kept secret.

District Court Judge William Blair Sylvester has ordered all motions by prosecutors and the defense be filed under seal so they cannot be inspected by the public. Sylvester also ordered the university not to release records on Holmes.

The news organizations include The Denver Post, three other newspapers, four Denver television stations, six broadcast networks and two newspaper groups.

A spokeswoman for the district attorney's office declined to comment. Holmes' attorneys didn't immediately return a call.

Meanwhile, authorities also are investigating whether Holmes practiced shooting in the remote mountains northwest of Denver.

The owner of C&M Guns in the town of Hot Sulphur Springs said local authorities, as well as agents with the FBI and the Bureau of Alcohol, Tobacco, Firearms and Explosives, have asked him whether Holmes came into his shop, which is near a public shooting range. Clark Branstetter said he didn't remember seeing Holmes.

KUSA-TV in Denver first reported the visit to that shop and another unnamed one in the area.

The public shooting range is owned by the Colorado Division of Wildlife. A spokesman wouldn't comment on whether Holmes used the range. The range isn't staffed and no records are kept of who uses it.

In late June, Holmes tried to join a private gun range east of Aurora but he never showed up.

As it turns out, those photos weren?t real; the images were actually of a 21-year-old Internet model who apparently has no qualms about taking her clothes off in front of the camera on a fairly regular basis. Catastrophe averted, right? Unfortunately, things only got worse from there.

On July 23rd, an alleged sex tape featuring the ?Call Me, Maybe? singer made its way online, prompting the Canadian pop star to address the leak on her Twitter page. ?Crazy morning. Discovered that someone put up a sex tape claiming to be me. Ridiculous,? she explained. ?Obviously not me.? Obviously.

Just when the nude photos and sex tape rumors had started to die down, another rears its ugly head. This time, however, the photos could be real. According to TMZ, the singer?s laptop was hacked not too long ago, and several personal images were taken as a result. As per usual, someone is attempting to shop the images to various websites, though no takers have been announced as of this writing. Authorities, meanwhile, are on the case, and if reports are telling the truth, they even have a suspect already in mind.

Considering the guy who hacked Scarlett Johansson?s phone is facing all sorts of trouble, I?m assuming Carly Rae Jepsen?s electronic intruder will face similar charges for his actions. However, here?s a good rule of thumb to live by: If you don?t want naked photos of yourself leaked onto the Internet, don?t take naked photos of yourself. Easy enough.

The same people who thirty years ago used to urge us to eat plenty of meat and cheese, shifted to encourage us towards vegetables and low fat salad dressings. And the tide is turning once again: there is a great deal of emphasis on the importance of good fats and essential fatty acids, most notably the Omega 3 kind. My point here is that it is foolish to establish absolute and narrow rules, especially about single ingredients in diets. Context counts.

A low fat diet is good for people who have eaten lots of meat and cheese and fat during their lives; it will help them re-balance and return to a more healthful equilibrium. On the other hand, a low fat diet is bad for people who have been eating raw food, fruit and vegetables for most of their lives. Not only that, a high fat diet is good for people with neurological disorders, particularly seizures. Seizures are not uncommon among young children, especially those who have been vaccinated.

Seizure disorder, or epilepsy, is ?an abnormality of the electric potentials or waves produced in the normal brain.? We fluctuate between the normal 9-to-14 alpha waves per second of the waking state and the 3-to-6 per second delta waves of the sleeping state. In epilepsy these waves go out of step, and an electro-encephalograph will indicate abnormal firings of the brain?s neurons.

Symptoms include grand mal seizures, with loss of consciousness and contraction and relaxation of all muscle groups, lasting a few minutes; petit mal seizures, which involve limited convulsions or temporary alteration of consciousness, sometimes extremely subtle; and psychomotor epilepsy, which consists of uncontrolled and unpremeditated behavior. In addition, related neurological disorders include severe headaches, absence seizures or staring episodes, dizzy spells, or impaired memory. Many of these conditions, incidentally, are common in criminals and juvenile delinquents1.

It is a fact little known by the general public that dietary modification can help seizure disorders. For example, the medical community has long been aware that fasting improves and even eliminates seizures. In the early 1920?s, R.M. Wilder introduced a diet intended to mimic the physiological effects of fasting. This diet consists of high amounts of fat (as much as 80% of calories) and low amounts of protein and carbohydrate (no more than 20% of calories total)2.

Like fasting, it provokes ketosis, the production of certain acids called ketone bodies which, in the absence of dietary carbohydrates, will be metabolized instead of glucose as fuel for the brain3; for this reason it was named the ketogenic diet (KD). Ketosis is demonstrated by testing the urine with dip-and-read sticks available in most drugstores.

From early on, the KD showed clinical effectiveness in suppressing and avoiding seizures. I had not heard about this until late 1994, when I read something about the ketogenic diet in a publication. When I discussed it in one of my classes, Carol Ellis, MD, a physician with an interest in alternative medicine, mentioned that she had studied the KD as a seizure controlling technique in medical school twenty five years earlier! Unfortunately, as it requires a great deal of effort to follow, and as even a small amount of extra carbohydrate can bring on seizures again within 3 1/2 hours, the KD was eventually neglected in favor of anticonvulsant drugs.

With the recent recognition that diet impacts health, interest in the ketogenic diet has returned. Johns Hopkins Medical Center has used it with great success to treat intractable seizures. In one review of 58 cases, seizure control improved in 67% of patients, and 75% of these improved patients continued the diet for at least 18 months. Sixty_four percent had AEDs (anti-epileptic drugs) reduced, 36% became more alert, and 23% had improved behavior . In St Louis Children?s Hospital it was found that 50% of patients on the KD, ranging in age from seven months to 38 years, had become free of seizures, and another 20 to 30% had experienced considerable improvement in control of their seizure5.

A version of the KD developed in Booth Hall Children?s Hospital in Manchester, Great Britain, requires that 50-70% of the calories be given as medium-chain triglycerides (MCT), a type of oil, plus 11% as other fats, plus 10% protein and 19% carbohydrate, a more palatable combination. The MCT oil is recommended for frying, grilling and in baked foods; it is also given as part of an emulsion to be sipped throughout meals, consisting of 200 ml MCT oil, 200 ml water, a teaspoon of powdered gelatin, and skim milk.

At first, this diet may provoke temporary abdominal pain, vomiting, or diarrhea, but rarely any other adverse effects; when effective, it may permit the reduction of anticonvulsant drugs without a return of the seizures. In a paper published in the October 2001 journal Pediatrics, a follow-up study of children with epilepsy found that ?Of the original 150 patient cohort, 20 (13%) were seizure-free and an additional 21 (14%) had a 90% to 99% decrease in their seizures. Twenty-nine were free of medications, and 28 were on only 1 medication; 15 remained on the diet.?6

Some researchers speculate that the high fat intake helps repair the myelin sheath around the nerves, a structure that is damaged in such widely disparate conditions as multiple sclerosis and adreno-leuko-dystrophy (the illness shown in the movie ?Lorenzo?s Oil?). Children with seizures need to remain very strictly on the diet, with not even the slightest deviation towards carbohydrates, for about two years. After that time they can slowly widen their food intake somewhat.

At a time when low fat is the defining factor for ?healthy? eating, I find it deliciously ironic that a high-fat diet could help with such a difficult problem as intractable seizures, and that it has been shown to be effective where drugs have failed. As someone once said, all food can be medicine, and all food can be poison; the secret is in using the right food at the right time. And there, my dear friends, lies the rub, and the reason for the many different prescriptions and recommendations in the dietary marketplace. It is essential that we keep an open mind.

How does one plan such a high fat diet? Following my inclination to use only real foods (not manufactured fats), I like what I found in a study from India. It includes the following daily foods used in the KD:

- butter and ghee (clarified butter) (6 oz)
- eggs (2)
- bread and chapati bread, (2 pieces)
- whole milk (10 oz)
- meat or dal (bean sauce) (2 oz)
- mixed vegetables (8 oz)
- orange (1).7

Breakfast: 1 egg fried in 1/2 oz butter, one slice toast with 1/2 oz butter, 4 oz whole milk.

Snack: 1 orange.

Lunch: 4 oz mixed vegetables saut?ed in 1 oz. butter, 1 oz meat with 1 oz melted butter.

Snack: 4 oz whole milk, 1 slice bread with 1/2 oz melted butter.

Dinner: 4 oz mixed vegetables saut?ed in 1 oz butter, 1 oz bean sauce with 1 oz ghee, 1 egg fried with 1/2 oz butter.

Random thoughts from a weight loss journey. ? jerseystrong

I have been doing a lot of reading lately about the problem of belly fat versus fat that is collected in other areas of the body. ?Belly fat is a more serious indicator of a potential health problem down the road. ? Heart disease and stroke being the number one risk from fat around the abdomen. ?The U.S. Army has even included waist circumference as a measure to predict healthy potential soldiers. ?The used to use BMI and body fat percentage solely.

Now, how do you specifically target belly fat. ?The standard answer that a lot of personal trainers will give you is that you can?t spot reduce fat. ?For example, if you have fat around your bicep than doing curls will not get rid of bicep fat. ?But, there is some good news for people who worry about this excess fat around the abdomen. ?If you read my blog you know that I am a runner. ?I am running the ING NYC Marathon this fall. ?I have also said that distance running is not a way to reduce body fat. ?Instead to reduce body fat high intensity sprints and power exercise such as full body olympic movements will help torch body fat. ?Well here is where it gets complicated. ?A recent Duke University study focused on cardio endurance training vs high intensity resistance training and their responses to visceral belly fat in particular. ?Visceral fat is the fat that sits deeper in your belly. ?It surrounds your organs and is much tougher to get rid of than subcutanous fat which is more superficial and lies just under your skin. ?The Duke study found that endurance training actually helped get rid of the visceral fat more so than high intensity training. ?This is why you can sometimes see a runner who has a very ?skinny frame? with a higher body fat percentage. ?If you look at them in a sweater you might say they are really skinny. ?However, if you asked them to take their shirt off you might see some belly fat. ?However, because their belly fat is that subcutaneous fat they are not at risk for heart disease etc. ?Now. think of the olympic weightlifter with a huge belly. ?This guy is powerful but he has the classic ?beer belly? look. ?He is at risk for a heart attack much more than the runner.

So, what does this mean for you. ?If long term health is your goal. ?I would try to amp up your cardio. ?When I began running I could barely run a mile. ?It?s just classic overload principal. ?Start slowly and add on a little at a time. ?Maybe add 10% a week at the most. ?When you first start don?t worry about distance just run for a time. ?Say 10 minutes at first. ?Then the next week 11 minutes. ?A good way to keep you motivated would be to sign up for a 5K or a 10K a few months down the road. ?These races are everywhere and they are a lot of fun with other people running alongside you. ?If you currently work out 4 days a week then maybe do 2 cardio days and 2 weight training days. ?When I was 290 pounds I went to the gym 6 days a week and lifted weights. ?I could bench press 340 pounds. ? ?But to work on that overall health and reduce fat you need to focus on diet and add cardio. ?Keep in mind however that I didn?t start running until after I lost 60 pounds or so. ?Diet is the key to all of this.

Lastly, but probably the most important is diet. ?When you are lifting weights you need protein to repair muscle tissue. ?However, if you trying to lose weight then you have to worry about total calories. ?I always say get to your ideal weight first then worry about shaping your body the way you want it. ?I use a smart phone app. ?There are a bunch out there. ?Loseit, MYFitnessPal, or FatSecret are all good. ?MyFitnessPal is my favorite. ?If you don?t have a smart phone they also have a website. ?Most people will tell you that eating at home is the best way to control weight because you can control the food that you prepare. ?However, with these calories trackers you can look up the calories in a menu item at say subway or Dunkin Donuts and make better choices.

This may sound obvious ?but many yo-yo dieters just don?t do this. ?EAT FOODS YOU LIKE! ?If you are chewing carrot sticks and you just hate them then how long do you think you are going to do that? ?Find lower calorie options that you actually like to eat. ?I can?t answer this for you but there has to be something out there that you enjoy that is not 1000 calories. ?If you don?t do this then your diet will be temporary. ?You want this to be a permanent change in the way you eat and not a diet.

Cheat Days are a must. ?There are things that everyone loves that they just can?t have when restricting calories. ?My cheat day is usually Saturday night and sometimes Sunday if I am training for a race. ?I will usually have a few beers and dessert on Saturday nights along with dinner.

Don?t diet at special events. ?Christmas is not a time to worry about your weight. ?If you start to live this way you will always be aware of calories and what the calorie count is. ?It will always be in the back of your head. ?If you feel that you have control over your diet then go ahead because you can always get back to your routine.

It takes 14 days to create a habit. ?I would recommend that when you start this new lifestyle that you are extremely strict for 2 weeks. ?No cheat days for at least 2 weeks. ?Your body will adjust and you won?t feel as hungry after two weeks.

Eat to not be hungry. ?Don?t eat until you are full. ?Think about this. ?I am a foodie. ?I love food but biologically we eat because we need energy. ?Our bodies signal hunger. ?Once that signal ends than we no longer need to eat.

Eat when your hungry. ?If you are hungry Eat! ?Usually just a small amount of food will actually end your hunger.

Successful dieters are boring. ?They tend to eat the same things over and over again. ?Because they know that it is something they like and they know the calories. ?It makes it easy and convenient.

Finally, crate your own Eat This not That. ?Men?s health popular program that lets you make small changes to your diet that will save you calories each time. ?This was probably my number one weapon for weight loss. ?For example, ?I stop at Dunkin Donuts everyday after work. ?I used to get an Iced Coffee and a muffin. ?I go from work as a school teacher right to my personal trainer job and needed something to hold me over until dinner. ?After learning that a muffin at Dunkin Donuts has 600 calories and a chocolate glazed donut has only 350. I switched to a donut. ?How many weight loss guys will tell you to eat donuts!? ?But I just saved myself 250 calories a day 5 days a week. ?In three weeks if I did nothing else I would lose a pound right there. ?After a few months of ordering glazed donuts I also like egg sandwiches. ?The Ham, Egg, and Cheese, on an english muffin is 290 calories. ?Bam! ?I just saved 60 more calories a day. ?3 months later they have an egg white flatbread turkey sausage sandwich. ?That is 210 calories. ?It is basically the same size and it fills me up just as much. ?I just saved 80 more calories a day. ?From my original muffin I am down 390 calories a day every day of the week. ?My original daily snack when I started going to Dunkin Donuts was an iced coffee with cream and sugar and a muffin. ?700 calories. ?Now I get am Iced Coffee with Skim Milk and splenda and a bacon, egg, and cheese wrap. ?for 215 calories. ?I did not make all of these changes at once. ?That is the key. ?I changed one thing and when I got used to it I then changed another thing. ?Think about your daily habits and how you can make small changes one at a time.

Coffee with no cream

No cheese on a sandwich. ?(would you really miss it) ?that?s 150 calories.

Mustard instead of Mayo

Diet soda instead of regular soda

Brown rice instead of white rice.

Weight watcher?s bread vs your regular bread.

Lite beer instead of regular beer.? etc

Find which swaps work best for you and remember choose one thing at a time.? All of these swaps for me took place over the course of several years.? If you tried it all at once then you would miss it too much because you are too close to your old habits.? Start with one swap and then let it become a habit.

Would a Dell Windows XP Installation CD work on a Custom made computer?

So as you may be aware, it is possible to automate the installation of a Windows operating system. Basically the "Dell" installation CD is really just Windows XP + specific device drivers embedded into the install process and possibly other Dell specific items. I suspect that you should be able to get through the installation minus device drivers.

With regard to the keys, Home edition and professional will have different keys. otherwise, people would buy the home edition and use the key on professional.

Gun shop veto draws legal fight

The latest location where the Second Amendment Foundation has taken its fight against unconstitutional limits on gun ownership is Alameda County, Calif., which changed the rules as three businessmen were trying to open a gun shop.

The Second Amendment Foundation was integral in the landmark McDonald v. Chicago Supreme Court case which determined that the Second Amendment provisions that individuals can have and own weapons applies to states.

That ruling followed a key decision in Washington, D.C., that earlier said the Second Amendment applied to individuals, not just militias.

Since then, the organization has taken on a number of gun restrictions set up by local governments, with the California location the latest flashpoint.

There, the foundation filed a lawsuit recently against the county and its board of supervisors for allegedly violating the constitutional rights of three businessmen by wrongfully denying them permits to open a gun shop.

The foundation was joined by the Calguns Foundation, California Association of Federal Firearms Licensees and businessmen John Teixeira, Steve Nobriga and Gary Gamaza.

They are represented by attorneys Donald Kilmer of San Jose and Jason Davis of Rancho Santa Margarita.

?The facts in this case are outrageous,? said SAF founder and Executive Vice President Alan Gottlieb. ?In the fall of 2010, Gamaza, Nobriga and Teixeira formed a business partnership with the intention of opening a gun store in Alameda County. When they began the process of getting permits to open their shop, they were advised of a requirement that gun stores not be located within 500 feet of any school, liquor store or residence.?

He said they carefully measured and found the location they had picked for their plans was ?well beyond the 500-foot limit.?

However, ?Then the county changed the measurement requirements,? he said.

The lawsuit explains the county also allowed an objection to the shop to be filed even though the deadline had passed, and even though the West County Board of Zoning Adjustments had voted to approve a conditional use permit and allow the gun store to operate.

?It is clear from the county board of supervisors? behavior that they have gone out of their way to prevent three businessmen from opening a gun store in their jurisdiction,? Gottlieb stated. ?This is a violation of their rights of equal protection and due process under the Fourteenth Amendment, and cannot be allowed to stand.?

He continued, ?Social bigotry is bad enough when practiced by the media and the gun prohibition lobby, but when it becomes the official policy of an elected government panel, it then becomes necessary, if not imperative, for the courts to intervene.?

The foundation also recently argued a pair of California cities and the state?s Department of Justice improperly confiscated firearms during investigations and then refused to return them to their owners ? even after the subjects of the inquiries were cleared.

That case is on behalf of gun owners Douglas Churchill and Peter Lau, and alleges the cities are engaging in ?deliberate theft of personal property.?

?We saw this sort of property theft following Hurricane Katrina,? Gottlieb recalled in a statement, ?and we took that case to federal court and won. Government agencies simply cannot seize private property and refuse to give it back by playing bureaucratic games.?

Gene Hoffman, chairman of the Calguns Foundation, which has also joined the case, explains, ?Law-abiding Californians should not be forced to seek out expensive legal representation just to get back what is rightfully theirs in the first place.?

According to court documents, Lau?s firearms were confiscated by the Oakland Police Department when authorities were investigating his brother?s suicide. Eventually, his guns were returned, all except one rifle the police deemed an ?assault weapon.? Lau?s attorneys dispute the classification.

Churchill?s firearms were confiscated by the San Francisco Police Department in January 2011 as part of an investigation, but the district attorney dismissed charges less than a month later. Nonetheless, police refuse to return seven of Churchill?s weapons ? including a Remington .22-caliber rifle and a Winchester 20-gauge shotgun.

?What the police departments are doing is a deliberate theft of personal property, and they know it,? added Gottlieb. ?Our partners at the Calguns Foundation have properly argued that this is inexcusable, and they are right.?

Names as defendants in the case are the cities of Oakland and San Francisco and their respective police departments, California Attorney General Kamala Harris and the state?s Department of Justice. The case is currently before the U.S. District Court of the Northern District of California.

Other cases the SAF has handled in recent months:

  • SAF sued the state of California over a ?vague? gun ban over a case in which a man twice was jailed and then cleared. The focal point is the definition of an ?assault weapon.? The statute?s definition of weapons is so ?vague and ambiguous,? the group contends, that one man was arrested on two different occasions for violations but ultimately cleared of any wrongdoing. ?It?s an insult to be arrested once for violating a law that is so vague and ambiguous that law enforcement officers cannot tell the difference between what is and what is not a legal firearm under this statute,? said Gottlieb, ?but to be arrested and jailed twice for the same offense is an outrage.?
  • In New York, the organization has asked for a summary judgment that would strike New York City?s $340 triennial fee for just owning a handgun. The legal brief explains that under U.S. Supreme Court rulings ?the right to keep a handgun in the home for self-defense is a part of the ?core? of the Second Amendment?s protections.? The case, brought by SAF, the New York State Rifle and Pistol Association and individuals including an electrical contractor, a paramedic, CPA and woodworker, argues, ?The city?s $340 fee is inherently prohibitive and serves the impermissible purpose of discouraging the exercise of constitutional rights. While the city can charge a nominal fee to defray costs, the $340 fee is not nominal, and has never been calculated to defray costs.?
  • The organization has sued New Jersey and officials and judges over procedures that allowed them to refuse firearms permits for a kidnap victim, a man who carries large amounts of cash for his business and a civilian FBI employee who fears attacks from radical Islamists. The permissions were denied on the grounds people had not shown a ?justifiable need.? ?Law-abiding New Jersey citizens have been arbitrarily deprived of their ability to defend themselves and their families for years under the state?s horribly crafted laws,? said an SAF spokesman. ?The law grants uncontrolled discretion to police chiefs and other public officials to deny license applications even in cases where the applicant has shown a clear and present danger exists.?
  • The SAF filed a case on behalf of an honorably discharged veteran from the Vietnam War and names as defendants Attorney General Eric Holder and the Federal Bureau of Investigation. The case was filed in U.S. District Court for the District of Columbia on behalf of Jefferson Wayne Schrader. The question is whether the state of Maryland can deprive an individual of the right to possess a weapon over a misdemeanor. Schrader had been convicted of misdemeanor assault relating to a fight involving a man who previously had assaulted him in Annapolis. But he was denied the opportunity to receive a shotgun as a gift or to purchase a handgun for personal protection.
  • SAF filed a claim against Maryland for a man who alleged the state was violating the Second Amendment by refusing to renew his handgun permit. Raymond Woollard originally was issued a carry permit after a man broke into his home during a family event in 2002. Woollard?s permit was renewed in 2005 after the defendant in the case was released from prison. But state officials later refused to renew the permit, even though the intruder now lives some three miles from Woollard.
  • SAF sued Westchester County, N.Y., because officials there were requiring that residents have a ?good cause? to ask for a handgun permit. The federal lawsuit alleges the requirement conflicts with the U.S. Supreme Court ruling that the Second Amendment establishes a personal right to ?keep and bear arms.? Individual plaintiffs in the case are Alan Kachalsky and Christina Nikolov, both Westchester County residents whose permit applications were denied.

Large, medically important class of proteins starts to yield its secrets

Readers of the top-ranked scientific journals Science and Nature might have noticed a recent wave of articles, most recently in the July 13, 2012 issue of Science, with deep importance for biology and medicine. These papers, all published this year by collaborations headed by the Scripps Research Institute laboratory of Professor Raymond Stevens, illuminate a large and medically important family of proteins called G protein-coupled receptors (GPCRs).

GPCRs sit in the cell membrane and sense various molecules outside the cell, including odors, hormones, neurotransmitters, and light. After binding these molecules, GPCRs trigger a specific response inside the cell. Many drugs, including allergy and heart medication and drugs for Parkinson's and Huntington's disease, target these proteins.

This year, a paper published January 19 (Liu et al., Science, 335, 1106) was quickly followed by related publications on the crystal structures of a lipid GPCR (Hanson et al., Science, 335, 851, February 17), the kappa opioid receptor (Wu et al., Nature, 485, 327, March 21), and the nociceptin opioid receptor (Thompson et al., Nature, 485, 395, May 17). The most recent publication is on the 1.8 angstrom high-resolution structure of the A2A adenosine receptor (Liu et al., Science, 336, 232, July 13) and is one of the highest resolution structures to date of a human membrane protein. The structure highlights the receptor and ligand as an allosteric machine controlled by sodium, water, cholesterol, and lipids.

These findings were made possible by technologies developed by the NIH Common Fund Joint Center for Innovative Membrane Technologies (JCIMPT) and the biological questions pursued by the GPCR Network, part of the Protein Structure Initiative:Biology at the National Institute for General Medical Sciences (NIGMS) in Bethesda, Maryland (NIGMS PSI:Biology).

Elusive GPCRs

The precise, three-dimensional arrangement of its constituent atoms is in some ways a protein's ultimate secret. Far more than its amino-acid sequence, the 3D structure holds the key to understanding how a protein interacts with its natural partner molecules in the body or with drug molecules.

But membrane protein structures are as hard to determine as they are valuable, and the most important structures, many of them GPCRs, have often been the most elusive. GPCRs are exceedingly flimsy, fragile proteins when not anchored within their native cell membranes. Coaxing them to line up to form crystals, so that their structures can be determined through X-ray crystallography, has been a formidable challenge for decades. For this reason, the NIH Common Fund highlighted membrane protein expression and stabilization technologies as a priority area of innovation and investment in 2004.

The first high-resolution human GPCR structure determined was the ?2 adrenergic receptor, published by Stevens' lab and the lab of Stanford's Brian Kobilka in 2007. In the next three years, only four more GPCR structures made it into the literature?and finding the structures of the several hundred other medically relevant GPCRs in the human proteome seemed a task for future generations.

Now there is much more optimism. "Ray's network is targeting GPCRs in such a way as to get representative structures from the various GPCR structure groupings, and for these we're probably going to need only about 100 structures," said Bryan Roth, a pharmacologist at the University of North Carolina whose lab collaborated with the GPCR Network to find the kappa opioid and nociceptin receptor structures. "The community may need only a few more years to do that, and of course, already we're finding structures for major GPCR drug targets, such as opioid receptors, that are going to have big impacts on future drug development."

The Technical Breakthroughs

What was the game-changing technical breakthrough?

"I'm always asked that question," Stevens said, "and the answer is that there wasn't just one breakthrough, there were about 15 separate developments, each one critically needed in combination with one another, and they came together after a long time. These are the results of 20 years of commitment by several international groups moving the field forward and the critical decision by the NIH Common Fund to focus on membrane protein technology development as a priority area of importance and investment for all of the NIH institutes."

Some of these breakthroughs have improved researchers' ability to produce and purify GPCRs in quantities sufficient for crystallization?a process akin to uranium enrichment. Other breakthroughs have been aimed at stabilizing GPCRs, whose core structure is made up of seven membrane-bound helical elements. "When you take away the membrane, these helices have the potential to fall apart," Stevens said. "It is possible that human GPCRs evolved to be unstable as part of their natural function to avoid over stimulation or signaling."

Over the past eight years, researchers with funding from the NIH Common Fund have developed and improved three key GPCR stabilization and crystallization techniques: the use of fusion proteins that stabilize the basic GPCR structure and make it more crystallizable without affecting its function (Chun et al., Structure, 20, 967, 2012); the use of drug compounds that bind to a GPCR and hold it in a specific functional conformation (Xu et al., Science, 332, 322, 2011); and the use of a membrane-mimicking matrix of fat and water molecules, called the lipidic cubic phase (LCP), in which GPCRs, cholesterol, and ligands can form crystals more readily than they do in traditional detergent solutions.

Improvements in these techniques have come by automating and expanding the LCP tool set. Stevens and Assistant Professor Vadim Cherezov built on the LCP work of structural biologist Martin D. Caffrey at the Trinity College Dublin, Ehud Landau at the University of Zurich, and Jurg Rosenbusch at the Biozentrum in Basel, Switzerland. "Stevens and Cherezov really advanced the access and more general use of LCP for crystallizing membrane proteins," said Jean Chin, the NIH program director for both the Common Fund JCIMPT and the GPCR Network.

"The group also developed precrystallization screening tools, next-generation crystallization robotics, imaging technologies, and working with staff at GM/CA beamlines at the Advanced Photon Source (APS) to develop technology to scan the LCP in order to locate the diffraction where the crystals are," said Ward Smith, director of the PSI:Biology program which along with the NIH Common Fund has provided funding for the work.

When GPCR crystals have been formed and isolated, they are sent to a special X-ray crystallography facility?often the Advanced Photon Source at Argonne National Laboratory in Argonne, Illinois. "At Argonne they've been able to get the X-ray beam smaller and smaller over the years," said Stevens, "and the beamline team at GMCA led by Janet Smith, Robert Fischetti, and Nukri Sanishvili have been incredibly creative and responsive to the needs of our research by developing such novel technologies as the rastering approach to find micro-crystals in the small x-ray beam with our samples as just one example."

Making Sense of the Data

Even when the crystal structure of a GPCR has been solved, scientists need to do further analyses to make full use of the structural data and receptor dynamics are central to their function. GPCRs typically function in part by binding to partner "ligand" molecules that are present outside the cell; that binding event may or may not initiate a biochemical signal inside the cell.

At Scripps Research, the laboratory of Kurt W?thrich, Nobel Laureate and the Cecil H. and Ida M. Green Professor of Structural Biology, applies nuclear magnetic resonance (NMR) techniques to determine how conformational equilibria and intramolecular rate processes in GPCRs contribute to their binding and signaling functions. "My lab has been working for years with Ray's, combining our NMR expertise with their crystallography expertise," W?thrich said.

In the paper that started the 2012 wave this past January in Science, for example, W?thrich's group published the NMR-based study of how different drug compounds affect signaling-related shape changes in the beta-2-adrenergic receptor. Such NMR structure studies were pioneered by W?thrichand one of the recent innovations has been the use of "microcoils" to enable NMR studies of minute quantities of precious protein samples in solution. W?thrich's lab, in collaboration with the Stevens lab, has also contributed innovative chemistry techniques for solubilizing GPCRs and other membrane proteins, both for crystallographic and NMR studies. "It's not trivial to make a good solution of a membrane protein," he noted.

The Networks

This quiet revolution in GPCR structure-function hunting has expanded because of the NIGMS's Protein Structure Initiative (PSI), which started in 2000. "Our basic idea was to apply some of the technologies that were enabling faster and cheaper gene sequencing to the problem of determining the structure of proteins and other macromolecules," Smith said. The initiative has been a success on the whole, but progress in solving membrane protein structures lagged at first. Smith noted, "When we renewed the effort in 2010, we decided to emphasize the funding of centers that would concentrate on particular membrane protein families."

The PSI program has been the catalyst in forming efficient structure-hunting research networks. The new NIGMS-supported "GPCR Network," centered at Scripps Research, includes: Stevens's lab; W?thrich's lab; the lab of LCP innovator Vadim Cherezov, an assistant professor of molecular biology; Professor Hugh Rosen, who leads the NIH Molecular Screening Center at Scripps Research; Professor Peter Kuhn, who is focused on x-ray advancements; Scripps Research Assistant Professor Vsevolod Katrich, who is focused on computational biology; and Professor Ruben Abagyan, University of California, San Diego (UCSD), who also focuses on computational approaches for the GPCR Network.

Some of the network's recent and ongoing projects have been initiated by outside labs that have special expertise in the biology of particular GPCRs. In fact, Smith says, the NIGMS uses its funding to encourage these collaborations. In 2009, for example, UCSD Professor Tracy Handel collaborated with Stevens and others in the GPCR Network to find the structure of CXCR4, an immune receptor on lymphocytes with relevance for HIV infection and the mobilizing of immune cells. The collaboration resulted in the publication of the CXCR4 structure in Science in 2010.

"The GPCR Network has amazing infrastructure and talent that allowed us to rapidly learn from their successes and delve into a challenging area of structural biology, which we could not have done without them," said Handel. "It was an incredible opportunity."

Another collaboration originated in 2008 when Roth, an opioid receptor expert, met Stevens at a conference. Over several years they worked on the problem of crystallizing the kappa opioid receptor. Technological developments from Scripps Research, such as the LCP innovations, made a big difference?as did Roth's suggestion of a stabilizing ligand, an experimental kappa opioid receptor antagonist called JDTic. Roth's lab also had funding from NIH's National Institute on Drug Abuse to automate post-crystallographic analysis.

"We were able to do a biological validation of the structural model in about three weeks, and that sort of thing used to take about a year," said Roth. "The automated protein engineering and crystallization techniques used in Ray's lab also speeded things up considerably."

Excitement About the Future

Currently only a few labs around the world have access to these new and highly automated technologies for structure determination, but that will change soon. "The NIH Common Fund JCIMPT has made a significant effort in outreach to transfer the technology to other groups interested in GPCR and membrane protein structural biology," Handel said. Roth added, "I think we're going to see a lot more GPCR structures being solved as these technologies are disseminated and these collaborative networks expand."

"We're in only the second year of the GPCR Network, but already the output has been tremendous; we're very excited about the future," said Stevens. "With the continued high impact of the investment in technology, we are only limited by outreach and resources in terms of the output and discoveries of the center. Perhaps most exciting is that we can finally start to address scientific molecular-level questions about GPCR evolution and perhaps even start to make links to human evolution and human cognition, the big picture questions that we are interested in pursuing."
